Ayşe Sultan was the daughter of Mihrimah Sultan, the daughter of Kanuni Sultan Süleyman, and Grand Vizier Rüstem Paşa.
Her first marriage was to Grand Vizier Semiz Ahmet Paşa, with whom she had four sons. Her second marriage was to Nişancı Feridun Paşa.
According to some reports, she later married Aziz Mahmud Hüdayi Efendi. In the later part of her life, she undertook a pilgrimage with her son and passed away upon her return at her palace in Üsküdar.
Known for her charitable acts, Ayşe Sultan commissioned a mosque in Üsküdar bearing her name (1594–95), the Celveti Lodge in İmrahor, a fountain, and a Qur’an recitation school (darülkurrâ).
Among her sons were individuals who held high state offices and were martyred in battle.

The tomb is one of nine tombs constructed adjacent to one another.
Between 1935 and 1940, the roofs collapsed and the structure was severely damaged.
Today, the interior has been reorganized with concrete sarcophagi.
In addition to Ayşe Sultan, her son Mehmet Paşa, daughter Ümmü Gülsüm Hanım, and her husband are also buried in the tomb.
In the past, the tomb was adorned with valuable calligraphic panels, oil lamps, carpets, and sarcophagus coverings.
Due to its location, it is one of the important burial grounds associated with the Ottoman dynasty in the vicinity of the Hüdayi Lodge.
